The large-scale film adaptation of Paul Gallico's novel begins on New Year's Eve on the luxury ocean liner Poseidon, sailing from America to Greece. The celebration is interrupted by the sudden impact of a giant wave caused by a nearby underwater earthquake. The water falls on the ship and turns it upside down.

A small handful of people, led by a priest, miraculously manage to get into compartments that are not yet filled with water. Passengers will have to endure several terrifying hours in an intense struggle for survival before they see the light of the sun again.
